Can there literally be four different positions to argue in a debate?
What does this all mean?

Replies - in some debate formats, after the main rounds are concluded, each side gives a summary speech that's half the length of the main speeches. The order is reversed ie whoever speaks first in the debate speaks last in the replies.
Rated - whether the site goes through a few dozen lines of math to recalculate your ELO score as a result of the debate.
Autoforfeit - in a 1v1 debate, you automatically win if the other guy forfeits
in a 2v2 debate, a debater that forfeits one round also forfeits all their remaining rounds
Filtering - allows only users/teams of a certain ELO to accept the debate
Can there be four positions? Kinda. Basically there are two teams that are pro and two that are con, but for obvious reasons the one pro team (for example) can't just copy the other pro team, but also can't contradict them. It's a very tactical kind of debating. See also: British Parliamentary.

A similar query, what does it mean to have no prep time? That you are given an on the spot topic or that the time to respond is less or something else?

Prep time is additional time after everyone has joined the debate but before the debate begins. It has nothing to do with the time to respond or any of that, but if it's a secret topic debate the topic is revealed. The purpose is to give everyone time to research the topic.
